A New simplified method for seed production of Edible Fungi
This method can eliminate a series of expensive equipment, raw materials, drugs and supplies needed for traditional seed production, and save thousands of yuan in investment. The strain produced has strong resistance, which is resistant to both high temperature and low temperature. The success rate of inoculation for propagation is 100%, and the seed production period is shortened by 20 to 30 days.
1. Innovation of raw materials for seed production
The simplified conventional method uses potato, glucose and Agar as raw materials to prepare PDA medium, which is used to transplant grade 1 test tube mother seed. The innovation takes one of the wheat kernels, corn kernels, corncobs and tree branches readily available to farmers as raw materials, and the universal medium of full-nutrition and high-nitrogen granular edible bacteria is made by coated with strain coating agent. it is used for grafting grade 1 test tube mother seed, grade 2 bottled original seed and grade 3 bottled cultivated species. The raw material cost of one test tube mother culture medium is only 0.03 yuan, and the raw material cost of one bottle of original seed or one bottle of cultivated seed is only 0.2 yuan, which is not only economical but also practical.
2. Innovation of high pressure sterilization
The simplified mother or original culture medium is put into a special high-pressure sterilizer with 1.2kg per square centimeter to maintain the sterilization method of 30~40min under steam pressure; the innovation is to put it into the "Subor" household pressure cooker for cooking, keep 40~50min when steam is ejected from the pressure-limiting valve hole and cool to atmospheric pressure and take out.
3. Innovation of atmospheric pressure sterilization
The simplified culture medium was put into the atmospheric pressure sterilizer for 8h, and the innovation was put into the space sterilization package (the cost of self-made material is about 30 yuan, which can hold 500m 800 bottles) for 3h.
4. Innovation of inoculation method
Simplify the old method of fumigation and sterilization with formaldehyde, potassium permanganate and other drugs in the inoculation box or inoculation room, and then use alcohol lamp flame to carry out closed inoculation; the innovation is to use a "four-no" sterile inoculator for edible fungus, without electricity, steam, alcohol or drug fumigation, and directly carry out full-process and open inoculation and transplantation of all kinds of edible fungi, with a finished product rate of more than 99%.
5. Innovation of clinker seed production
The third-grade cultivation production of edible mushrooms such as Pleurotus ostreatus, Coprinus comatus and Pleurotus ostreatus was simplified and inoculated with "clinker" after steam atmospheric pressure sterilization. After loading, the stomata were perforated in the middle, and the granular species with full nutrition and high nitrogen were connected (15% of the seeds) and cultured at the right temperature.
