Then put the mouth of the test tube that needs to be stored on top of the alcohol lamp for rotary sterilization. Then take out the wax paper and fasten the mouth of the pipe tightly. Then put the mouth of the tube into the melted paraffin and wrap it tightly in wax paper to avoid air leakage. After the paraffin has cooled, store it in the refrigerator.
3. Cellophane seal storage
Cellophane sealing storage first needs to prepare the sealing agent, with gelatin, copper sulfate, methylene blue and other items into hot water to dissolve and mix together. Then pull out the cotton plug of the test tube full of mother seed, face down, and seal it with sealing agent. Then put the cellophane in and soak it, then stick it to the mouth of the tube and dissolve it with paraffin wax. At room temperature, this storage method can last at least 8 months, and can avoid the pollution of pathogens and pests.
4. Barnyard manure storage
Barnyard manure storage is very suitable for the storage of straw rot fungi such as Pleurotus ostreatus. First of all, the rotten cow dung should be dried and broken, then sifted with a sieve, add an appropriate amount of water to adjust the humidity, put it into the test tube, and keep the depth at about half. Then carry on the sterilization work to ensure the inoculation after asepsis. When the mycelium is full and stored in the environment of about three degrees, the storage time is very long, and its production characters will not change greatly.
