Firstly, the high quality mother seed was made, and the mother seed culture medium was prepared at the same time. Finally, the cultivated seed is made and can be sown after it grows all over the bottle wall.

1. Mother culture medium formula: potato 200g, glucose 20g, Agar ZO g, potassium dihydrogen phosphate 1g, magnesium sulfate 0.50g, water 1000 ml PH 5.50g 6.50g.

Inoculation culture: after inoculation according to the conventional method, the mother seed was put into the culture room at 23: 25 ℃, the air relative humidity was about 65% for 7 days and 10 days, and the hyphae covered the slope of the culture medium, which could be used to pick up the original seed.

2. The original formula: wheat (corn): 95%; gypsum powder: 20% calcium superphosphate: 20%; urea: 0.50%; white sugar: 0.50%; pH value: 5.50% 6.50. 2.1.2 seed hull: 87%; wheat bran: 10%; sucrose sugar: 0.50%; gypsum powder: 1%; calcium superphosphate: 1%; urea: 0.50%; water 120%: 130%.

Inoculation culture: when the culture medium of bacteria A was cooled to room temperature, each test tube could be inoculated with 5-6 bottles of original seeds. The culture conditions are the same as those of the mother species, and the bottles can be filled in 20 to 25 days. Note: shake the bottle after 10-12 days of culture, and then culture it. The characteristic of high-quality original species is that the hyphae are white and neat, the walls and surfaces of the bottle are covered with mycelium, and there is mushroom flavor.

3. Production of cultivated species

Inoculation culture: first, wipe hands with 75% alcohol or 3% laisuer and disinfect the original seed outside the bottle, then use a sterilized inoculation shovel (hoe * to loosen the original seed, pour a little of the original seed into the sterilized bottle above the alcohol lamp flame in the inoculation box (on the super-clean workbench), each bottle of the original seed is connected to 20 bottles, and the culture conditions are the same as the original seed. Generally, it can be full in 20 to 25 days, and should be sown immediately after it is full. If it is not used temporarily, it can be dried and preserved at 10-14 degrees Celsius for no more than 10 days, and no more than 20 days at 2-4 degrees Celsius. The cultivated species preserved at low temperature were restored at room temperature for 1-2 days before use.
