Edible fungi have rich nutritional value, low production investment and high economic benefits. In recent years, there has been an upsurge in the production of edible fungi in rural areas. the mycelium of edible fungi propagates rapidly and is easy to aging. The conditions in rural areas are limited, and it is more difficult to preserve bacteria. Here are seven simple and easy preservation methods for agricultural products:

The distilled water method is suitable for the seedlings of Pleurotus ostreatus, Flammulina velutipes and Lentinus edodes.

1. After the preservation of Pleurotus ostreatus and Flammulina velutipes, 80% of the bacteria to be preserved in the test tube were transferred into the inoculation box for sterilization, and distilled water was injected into the test tube plug with a syringe to submerge the culture medium to 2 cm at the mouth of the tube. The mouth of the tube is sealed with mineral wax and stored in a cool place at room temperature for 1-1.5 years.

two。 The strains of Lentinus edodes and Auricularia auricula were preserved. Under the aseptic operation in the inoculation box, the solid or semi-solid test tube cultured to medium well was put into a large test tube of 25mm × 200mm. The large test tube was injected with distilled water to 1 cm at the mouth of the tube, sealed with rubber bolt, strengthened and sealed with mineral wax and placed in a cool place. Auricularia auricula can be preserved for 7-8 months and Lentinus edodes for 9-12 months.

This method can also preserve other bacteria at natural temperature.

Second, the urea temporary storage method places bagged urea in a place that is dark and well ventilated, the main body of the test tube bacteria is buried in the urea particles, and the mouth of the test tube is exposed, which can preserve the vitality of the bacteria for as much as 30 days. This method can be carried out in a constant low temperature environment.

3. Liquid paraffin preservation method: chemical liquid paraffin is selected. After high pressure sterilization, it is put into a 40 ℃ thermostat for several hours or in a dryer for several days, and liquid paraffin is absorbed by a sterile straw or syringe and injected into the strain test tube. The injection volume should be about 1 cm higher than the tip of the bevel, plug the pipe mouth with cotton plugs and seal with paraffin wax. Store upright in a clean, dry, cool place for 5 years.

Fourth, bran preservation method to take a certain amount of wheat bran with water or nutrient solution, mix well and then set aside. The water bran was put into the test tube and sterilized under high pressure, then mixed with the bacteria, cultured at the appropriate temperature, and dried at room temperature after the mycelium developed. Then store it in a dry place below 20 ℃, which can be preserved for 3 to 5 years.

5. The bacteria were inserted into the potato culture medium by normal saline preservation method. The 60ml culture medium was filled in a 250ml triangle bottle, and the culture medium was shaken 5 times a day or 10 times a day for 5 days. Then the mycelium balls formed were inhaled into the test tube containing 5ml sterile saline, and about 4 to 5 mycelium balls were transferred into each tube. The low temperature can be preserved for 1 year.

Sixth, wheat grain preservation method to take high-quality wheat after washing, soak in 20 degrees Celsius water for 5 hours, slightly dry and then tube. The appropriate loading quantity is from 1 stroke 4 to 1 stroke 3 of tube length. The test tube containing wheat grains was sterilized and dispersed while it was hot, cooled and inserted into the mycelium. The culture was terminated when sparse hyphae appeared on most wheat grains at the appropriate temperature. The strain can be stored in a dry place below 25 degrees Celsius for 1 to 2 years.

7. The refrigerator preservation method puts the test-tube bacteria that have been grown in the refrigerator. It can be stored in the fresh-keeping layer for 7 months. The strain does not degenerate.

The above methods are suitable for test-tube bacteria.
