Propagation technique of virus-free test-tube plantlets of potato

Through the smooth implementation of the poverty alleviation and development project of "hospital and county co-construction" and under the guidance of experts and professors of the Provincial Academy of Agricultural Sciences, Huachuan County has established a virus-free potato breeding base based on Sujiadian Town and Lifeng Township, which has greatly improved the cultivation technology level, yield and quality level of potatoes in our county. The average yield per mu is about 2000 kg and the starch content is about 20%. In order to further improve the production level of virus-free seed potato, we studied the propagation technology of virus-free potato test-tube plantlets. The main techniques are as follows:

1. Selection of non-toxic seedlings

Non-toxic test-tube plantlets used for propagation must undergo strict virus detection. It does not contain any viruses or viroids. Non-toxic test-tube plantlets with strong growth, 5-6 cm plant height, medium Internode, stem rent and large leaves.

two。 Preparation of culture medium

MS medium was used for propagation of non-toxic test-tube plantlets. Add the right amount of Agar. Due to the different contents of Agar sold in the market, experiments must be carried out before preparation. The dosage is about 613 grams / liter. Boil the culture medium and pour it into the test tube while it is hot, each tube is filled with 6-7 ml, plug with cotton plug and put into the sterilization pot. Sterilization pressure of 15lb (121lb) for 20 minutes can completely kill bacteria. After sterilization, it is taken out and cooled, and after solidification, it can be used for propagation.

3. Propagation equipment

Conditional units can use aseptic console, we focus on exploring the use of simple inoculation box for propagation technology. In addition to the operating box, you also need an alcohol lamp, planting hooks, iron plates, matches and alcohol cups containing 1Accord 3.

4. Disinfection

The day before propagation, the room was thoroughly disinfected with 3-5% Lysol, and the disinfection box was cleaned with 75% alcohol before disinfection. The sterilized medium equivalent to 5-6 times of non-toxic seedlings (each propagation) was taken and put into the operation box after disinfection. Disinfect the inside and outside of the operation box again.

After cutting, use the planting hook to insert the segments of each tube upright (non-inverted) into the medium. After planting, put the mouth of the test tube on the flame while burning and stuffing with cotton plugs.

After all the planting, the empty tube and the newly planted test tube plantlets were taken out.

5. Cultivate

The planted test-tube plantlets should be placed at room temperature 2224 ℃, light 1000~3000LUX, to ensure 16 hours of light every day, the lack of sunlight can be supplemented by fluorescent lamps. Under normal circumstances, it takes about 3 to 5 days to take root, and the next propagation can be carried out after about 25 days.

By using the above techniques, the number of non-toxic test-tube plantlets can be increased by nearly 700 times in 3 months in winter, and the production cycle of non-toxic seed potato can be greatly shortened.
