Preservation of Edible Fungi
1. Distilled water method. Suitable for mushroom, needle mushroom, mushroom strains.
1. Preserved mushroom and needle mushroom strains. First, transfer the 80 percent mature test tube strain to be preserved into an inoculation box for sterilization, inject distilled water from the test tube stopper with a syringe, and completely submerge the culture medium surface until the nozzle is 2 cm. The nozzle is sealed with mineral wax and can be stored for 1~1.5 years in a cool place at normal temperature.
2. Preservation of mushroom and agaric strains. Under aseptic operation in the inoculation box, put the solid or semisolid test tube strain cultured to seven-maturity into a large test tube of 25×200 mm with a finger tube of 150×15 mm, inject distilled water into the large test tube until the nozzle is 1 cm away, wrap the seal with a rubber plug, reinforce and seal with mineral wax, and place in a cool place. Fungus strains can be preserved for 7~8 months, mushroom strains can be preserved for 9~12 months.
This method can also preserve other strains at natural temperatures.
2. Urea temporary storage method. Put the bagged urea in a dry, well-ventilated place away from light, bury the main body of the test tube strain in urea granules, and expose the mouth of the test tube to the outside, which can preserve the activity of the strain for as long as 30 days. This method can be carried out in a constant low temperature environment.
Liquid paraffin preservation. Choose chemical liquid paraffin. After autoclaving, put it into a 40℃ incubator for several hours or a desiccator for several days, and inject liquid paraffin into the culture tube with a sterile pipette or syringe. The injection volume should be about 1 cm higher than the tip of the bevel, and the nozzle should be plugged with cotton and sealed with paraffin. It can be stored upright for 5 years in a clean, dry, cool place.
4. Wheat grain preservation method. Take high-quality @#@ 245 @#@, wash, soak in water at 20℃ for 5 hours, dry slightly, and install tubes. The loading amount should be 1/4~1/3 of the pipe length. Sterilize the test tube containing the wheat kernels and shake them hot. After cooling, inoculate mycelium. Culturing at appropriate temperature until sparse hyphae appeared on most wheat grains, terminating the culture. Strain can be stored in dry place below 25℃ for 1~2 years.

Preservation of edible mushroom strains
Usually take the following two methods of preservation: slope low temperature preservation method. This is the simplest preservation method, that is, the vigorous and pollution-free slant test tube is put in the refrigerator and preserved at a low temperature of 4 ℃ ~ 6 ℃, and then transferred to the fresh slant medium every 2 to 3 months, and then preserved after culture.
