The following problems should be paid attention to in the process of culture material preparation.

1 the site mixing and bagging site should be treated with cement, with a slope of 1%, so that the washing water can flow away naturally, rinse with clean water after daily operation, and clean the remaining culture materials for no longer use, so as to prevent the microorganisms in the remaining materials from entering the fresh culture materials, speeding up the rate of rancidity of the culture materials and increasing the chance of pollution.

(2) the sterilized culture material should be sterilized while mixing the material and bagging at the same time, the time from mixing to sterilization should not exceed 4 hours, and the temperature should be raised by fierce fire when loading the pot to sterilize, so that the culture material can enter the aseptic state as soon as possible.

(3) due to the difference of raw material water content and physical properties, the air temperature and relative humidity are different, so the water transfer must be flexible, when the culture material is dry, the particles are fine and the acidity is strong, the water can be adjusted more; the culture material has more water content, coarse particles and poor water absorption, and the water should be adjusted less. In sunny days, the bagging time is long, the water transfer is more or in the middle; on cloudy days, the air relative humidity is high, the moisture is not easy to evaporate, and the water transfer is less. The raw materials such as bagasse, corncob and cottonseed shell are loose, large and easy to absorb water, so the amount of water transfer should be increased appropriately.

(5) when the bagging time and temperature is on the high side, the bagging time should not be too long, which requires organizing manpower and racing against time to complete, so as to prevent the rancidity and nutrition of the culture material from decreasing. (6) in the preparation of bagged aseptic culture materials, in order to avoid pollution, on the basis of selecting good raw materials, mix materials in a sunny morning, and try to complete and enter the aseptic process in the morning with low temperature, so as to reduce the chance of miscellaneous bacteria pollution.
