The crude extract of Schisandra chinensis and its active component Schisandrin A can reverse multidrug resistance and greatly improve the sensitivity of tumor cells to antineoplastic drugs. The structure of Schisandrin An is different from the known multidrug resistance reversal agents, so it is a kind of Chinese herbal medicine with development prospect. This is a "new discovery" disclosed in his academic report by Liu Gengtao, a researcher at the Institute of Medicine of the Chinese Academy of Medical Sciences and academician of the Chinese Academy of Engineering, at the recent Guangzhou Pharmaceutical 2006 Cross-Strait Pharmacy (Yangcheng) Forum.

Liu Gengtao pointed out that cancer is one of the biggest killers of human health, and its mortality rate ranks first in China and ranks second in the world. Multidrug resistance of tumor cells is one of the important reasons for the failure of tumor chemotherapy. The so-called "multidrug resistance" refers to the congenital existence of tumor cells or induced by chemotherapeutic drugs to a series of antineoplastic drugs with different structures and mechanisms. Therefore, it is of great clinical value to reverse the multidrug resistance of tumor cells and restore the sensitivity of tumor cells to chemotherapeutic agents.

Academician Liu carried out a series of studies on "reversing drug resistance" of Schisandra chinensis in vitro, nude mice and anti-tumor experiments, which confirmed that the crude extract of Schisandra chinensis and its active component Schisandrin A had obvious effect on reversing multidrug resistance. In the experiment of enhancing the inhibitory effect of the crude extract of Schisandra chinensis and these three drugs on the growth of sarcoma S180 in mice, it was found that the anti-tumor effects of the three drugs were enhanced to a certain extent, and the tumor inhibition rates were 51.66%, 38.11% and 53.05%, respectively. In the mechanism study, Academician Liu found four mechanisms: first, Schisandra chinensis can increase the accumulation of antineoplastic drugs in drug-resistant tumor cells, thus enhancing the curative effect; second, it can inhibit the expression of P-glycoprotein, suggesting that it can play its role at least partly by interfering with the drug efflux pump mediated by P-glycoprotein directly or indirectly; third, it can significantly increase the susceptibility to apoptosis of hepatocellular carcinoma BEL7402 cells induced by adriamycin. Fourth, calcium antagonist VER can partially antagonize the increase of intracellular calcium induced by dihydropyridine calcium agonist BAYK8644, suggesting that Schisandrin itself has no calcium antagonistic activity.

The above studies show that the mechanism of Schisandra chinensis crude extract reversing multidrug resistance is mainly related to the inhibition of P-glycoprotein and the enhancement of antineoplastic drugs to induce apoptosis of drug-resistant tumor cells. Liu Gengtao said that the research results have now applied for patent protection. He believes that Schisandra chinensis is rich in domestic resources and is worthy of further research and development.
