Tissue Culture and cultivation Management techniques of Rhododendron

Mainly because most of the soils in North China are alkaline, and rhododendrons like typical acidic soils, so they have some difficulties in reproduction and planting, so they can only be used as greenhouse flowers. There are several kinds of rhododendron reproduction, such as sowing, cutting, striping and grafting, tissue culture and so on. Tissue culture does not need seeds, just use the stem tips of the current-year branches of rhododendron. A large number of small rhododendron seedlings can be cultured in test tube in a very short time, and it is not limited by the season.

Methods: first, take the top bud of the young branch of rhododendron, rinse it under tap water, then put it in a beaker, then pour into the washing solution, add appropriate amount of water, stir with a small brush, repeat twice, then rinse clean with clean water, put the washed stem tip into 75% alcohol and soak 1min, rinse it with sterile water for 4 times in a sterile super purification table, then soak 3min with 0.1% mercury. Then rinse it with aseptic water for 4 times and finally inoculate it into the differentiation medium. The differentiation medium was modified MS medium, in which KI was removed and the ratio of NH4+ to NO3- was adjusted from 1 ∶ 2 to 1 ∶ 1. The pH value was between 5.0 and 6.5.

MS (improved) + 6BA1.0-2.0 (mg/l) + NAA (0.1~0.5mg/l)

Clustered buds were differentiated on the stem tip after 1 month. After 20 days, the tufted buds were cut and inoculated on the rooting medium. After 15 days, the white young roots were formed. After 20 days, the root length was 2~3cm. The rooting medium was:

MS+IBA1.0~2.0mg/l+NAA0.1~0.5mg/l when the cuckoo seedlings grow in the test tube for about 20 days and the root length is about 2~3cm, the test tube seedlings can be moved into the greenhouse to make the test tube seedlings adapt to the external environment. When the test tube seedlings are refined in the field for about 4 to 5 days, the cuckoo seedlings are gently removed from the test tube and placed at a water temperature of 25 ℃ and a pH value of 5.0. Rinse the Agar from the fine root in 0.1% carbendazim solution

Then it was planted in a mixture of humus soil, moss and mountain mud with a mixture of 2 ∶ 1 ∶ 7. Potted plants in the greenhouse should pay attention to drainage, watering, spraying and other work. The seedlings transplanted on the same day should be fertilized once, and pay attention to light rather than thick, because the roots of rhododendron are very fine, and thick fertilizer is easy to rot the roots. In Heze area, most of the water is alkaline, so we should pour alum fertilizer water at the right time. Alum fertilizer water is made of 3kg ferrous sulfate, sesame oil residue 5kg, bean cake 5kg and water 200kg, which must be diluted and poured after ripening. When rhododendron blossoms, it is required to increase nitrogen fertilizer and shade properly in summer.
