Amoebas like to live in pools where the water quality is relatively clear or in shallow water where the flow is slow and algae are abundant. Sometimes it is attached to rotten plants submerged in water or mud, or on foam floating on the surface. Take the soil or rotten organic matter from the bottom of the pond and collect it with water and place it in a bottle for about 24 hours. This provenance was taken during culture. The optimum living temperature of amoeba is 18 ℃ ~ 22 ℃, which can be easily collected in spring and autumn. The amoeba is large and a white spot can be seen with the naked eye. Therefore, it can be separated as a provenance by making use of its characteristic that it is suddenly shocked and firmly attached to the object when it is full of food. The method is to drop the culture medium containing amoebas on the glass sheet, and the body will be firmly attached to the glass sheet, and then slowly rinse the culture medium on the glass sheet with cold water for about 10 seconds, so that several pieces can be made as provenance and put together with the glass sheet in the culture medium. After a few days of culture, a large number of pure amoebas can be obtained.
