1. Visual appearance method. The shape and color of maize seed embryos were observed with the naked eye. Where the seed embryo protrudes or shrinks, showing dark and no luster, the seed is fresh and has strong vitality and can be used for production.

2. Seed soaking and sprouting. First, soak 100 seeds in water for about two hours, put them on moist papyrus, cover with moist papyrus, and place them in an environment with sufficient oxygen and room temperature 10 Murray 20 ℃, so that the seeds can germinate fully; then divide the number of germinated seeds by 100, multiply by 100%, and obtain the germination rate. Although this method is accurate, it takes 8 days.

3. Red ink staining. The dye was prepared with a commercial red ink and 19 parts of tap water; 100 corn seeds were randomly selected and soaked in water for two hours to let them absorb; the inflated embryos and milk embryos were stripped out one by one with tweezers; the treated seeds were evenly placed in the incubator and injected with dye to drown the seeds. After dyeing for 15 minutes, the dye was poured out and the seeds were washed repeatedly with tap water. The dead seed embryo and endosperm are dark red, and the living seed embryo is not dyed or slightly reddish, according to which the number of living seeds is judged, divided by 100, multiplied by 100%, the germination rate is determined.
