Under suitable moisture and temperature conditions, the bacteria that were not killed in the bacteria bag began to reproduce, and began to distribute on the bacteria bag in spots, and later expanded into pieces. So what are the reasons for incomplete sterilization?

3. No colonization of strains or slow growth of mycelium

There are many reasons for this phenomenon, do not rule out some. Weak strain activity can cause hyphae to grow slowly. But most of the reasons were that the bag was too tight or the water content was too high (more than 65%), and the growth rate of hyphae was inhibited because there was not enough oxygen in the bag. Of course, too low a temperature, and poor ventilation or culture medium pH is not appropriate, will also cause slow hair bacteria. After sterilization of bacteria bag, the material bag is not fully cooled, and it is eager to inoculate, which leads to the phenomenon of non-colonization of bacteria. There is also a culture room is too dry, the material surface water loss is serious, and it is impossible to colonize the bacteria.
