Culture preparation

1. Mother species

Common media are PDA or PSA. MGYA medium (1000ml for example: peptone 1g, maltose 20g, yeast 2g, agar 20g), PDYA medium (1000ml for example: peptone or soybean peptone 1g, potato 300g, agar 20g, glucose 20g, yeast 2g), malt extract medium (1000ml for example: malt extract 20g, agar 15g, peptone 5 g) can also be used. According to the conventional method, it takes 8~10 days for hyphae to grow completely in tube. The growth speed of hyphae can be accelerated by using the last three media.

2. Original and cultivated species

The culture medium formula is the same, but sawdust bran culture medium can be adopted, and its formula is sawdust 73%, bran 25%, sugar 1%, and calcium carbonate 1%. Grain such as wheat, barley, corn and sorghum can also be used as culture medium. According to the conventional method, ingredients, treatment, bottling (bag), sterilization, inoculation, culture, the first medium generally takes 30-35 days to grow hyphae full bottle (bag), with the latter several grain media, hyphae full bag (bottle) faster, about 20 days.
