There are two methods of orchid seed germination, namely symbiotic germination and non-symbiotic germination. At present, the research on orchid seed germination is mainly focused on non-symbiotic germination, through this method, the seeds of Cymbidium, Jian Orchid, Chunlan, Cold Orchid and Cymbidium can germinate. But generally speaking, the germination rate of orchid seeds is low, it is difficult to regenerate plants, and the survival rate of test-tube plantlets is not high.
The orchid seeds are small and powdery. Under a microscope, the seed is shaped like a shuttle and consists of a transparent seed coat and a spherical embryo. There is no endosperm in the seed. The color of mature seeds is yellow and that of immature seeds is white. Although orchid seeds are small, they are numerous, and each capsule contains tens of thousands to hundreds of thousands of seeds.
It is better to sow orchid seeds with immature seeds. The sowing method is as follows: take the uncracked capsule, clean the stalk and other residues, rinse clean with running water; then disinfect with 75% alcohol for 8 minutes for 10 minutes and rinse with sterile water for 3 times; after the filter paper is dried, cut it with a scalpel and sow the seeds on the surface of the culture medium with tweezers; the inoculated seeds are cultured in the culture room. The culture conditions are as follows: the temperature is about 25 ℃, and the protocorms are cultured in the dark or under scattered light before the protocorms are formed.
